科研简介

科研哲学是揭示复杂现象中的简单美,服务全民健康。研究方向聚焦传染病计算系统生物学,开展相关基础与应用研究,侧重综合运用统计学习、理论建模以及高通量实验技术等手段,整合从微观到宏观的多维异质信息,定量研究多种系统因素与传染病的产生、演化、传播、感染免疫以及致病的复杂关系,揭示并定量描述背后隐藏规律及其机制,指导传染病日常监测、预警、防控与治疗。代表性成果包括基于序列流感快速准确抗原预测与疫苗株推荐,融合多维信息的季节性流感长期预测,融合统计建模与机制建模的传染病复杂场景模拟预测与评估,基于突变网络框架的病原体演化与传播分析,以及复杂感染免疫表型的系统生物学机制解析与应用等,研究成果先后在Science Translational Medicine, Nature Communications, Genome Research, Nucleic Acids Research, Briefings in Bioinformatics, PLoS Pathogen, Journal of Virology and Emerging Microbes & Infections等高水平杂志上发表。课题组公众号DuLab计算生物学

代表性成果

本课题组研究方向聚焦传染病系统交叉研究,针对目前传染病防控被动局面,融合微观演化、宏观传播与人群免疫信息,综合利用人工智能、数学建模与多组学技术,推进变异规律解析与预测、流行传播规律解析与预测以及人群免疫力定量解析与应用,辅助传染病精准科学防控。

变异规律解析与预测:病原体优势变异株预测对于精准传染病预测预警与防控至关重要,然而,由于我们对其影响因素及其规律缺乏全面理解,导致流行株及其对流行传播影响效果预测受限。病原体微观分子变异受自身蛋白结构及功能约束,同时也受到宿主免疫系统选择,在群体层面也存在竞争与淘汰,因此,要想有效识别优势变异株或预测变异方向,需综合考虑这些系统影响因素,了解清楚其中规律。从变异预测相关的微观、宏观及免疫系统影响因素的角度出发,发展创新理论方法来推进传染病病原体变异预测是本课题组其中的一个重要的研究方向,目前已在Nature Communications,Journal of Infections, Briefings in Bioinformatics等发表了相应成果。

 

传播规律解析与预测:传染病的流行传播受众多因素的影响,包括病原体本身变化、环境及群体因素以及宿主免疫规律等。传染病规律解析与模拟预测取决于影响因素与影响方式的有效识别,并在此基础上构建合适的理论模型,以实现有效模拟与预测预警。目前数据驱动的统计学习方法和机制驱动的数学建模方法在传染病建模方面都有广泛应用。本课题组致力于通过统计学习方法不断推进传染病流行传播影响因素及影响方式的有效解析,并在此基础上不断升级动力学机制建模以及整合统计人工智能和物理机制约束的融合建模以实现真实场景复杂传染病流行传播长效预测。目前该方向的相关成果已发表在Science Translational Medicine,Emerging Microbes & Infections,PLoS Pathogen等。

 

人群免疫力定量解析:传染病变异及传播必然受宿主免疫系统影响,同时宿主系统是一个庞大复杂的生物系统,因此传染病的有效防控需要考虑宿主本身复杂互作系统以及病原体与宿主互作系统。这方面的深入研究也将对药物设计、疫苗设计、疫苗策略及防控策略设计提供机制理论指引。本课题组致力于基于复杂系统解构视角推进传染病相关人群免疫力数字解码,目前已构建了自然健康人群抗体库跟踪队列以及免疫力跟踪队列,从宿主基因组、转录组和抗体库等高通量组学角度开展研究与应用。相应成果已发表在Emerging Microbes & Infections,Briefings in Bioinformatics,Frontiers in Immunology等。
